A group of overweight people are entered into a weight loss/dance competition. Housed together for the duration of their time in the competition, the contestants are provided with a general fitness trainer, a nutritionist, a physician, and a costume supervisor. Each is also provided a professional dance partner/choreographer, with who he/she performs a series of dance routines in front of a panel of dance experts, who score the routines on a scale of 1 to 10. Each week, the averaged dance score for each contestant is added to his/her weight loss score (the percentage of weight loss for that week). The person with the lowest overall score for the week is eliminated from the competition until there is one contestant left standing, who is named Dance Your Ass Off champion.

Head judge Lisa Ann travels from Los Angeles to Dallas to Chicago to New York to hold open auditions to find the twenty-five finalists to be called back to Los Angeles for a final audition to choose the twelve season 2 contestants. After all the applicants perform a choreographed dance routine and a solo dance, Lisa Ann and a different choreographer in each of the four cities choose the first eighteen finalists. Lisa Ann chooses the remaining seven from the hundreds of on-line submissions. In Los Angeles, the twenty-five are assessed by consulting physician Dr. Geller, and perform another dance audition - which includes both a choreographed group dance as well as a solo component - in front of judges Lisa Ann and Danny. From each of the two dance groups, Lisa Ann and Danny choose one applicant apiece to make it through to the show. After conferring, Lisa Ann and Danny request seven of the remaining applicants to perform one more dance routine. Lisa Ann, Danny and Dr. Geller choose the final eight based on dance ability, dance potential and current health and weight conditions which may impact their ability to perform the requested dance routines. Host Mel B. then announces those final eight contestants.
