"O. Henry's Full House" is a film divided into five segments telling five tales at the beginning of the 20th century. 1) "The Cop and the Anthem": Winter is coming and homeless drifter Soapy wants to go to jail for three months to get shelter and food. His partner Horace suggests that they look for shelter with the Salvation Army, but Soapy refuses. He forces many situations to be arrested, but he is always forgiven. When he goes to the church, there is a miracle and Soapy decides to seek a job position. Will he succeed? 2) "The Clarion Call": When a thief kills a man, the police investigators do not have any lead to follow. Police Sergeant Barney Woods sees a pen that was found in the crime scene and he seeks out a man called Johnny Kernan. He finds Johnny, who invites Barney to drink with him, and they go to his hotel room. Johnny recalls their youth, when they were friends, but Barney tells him that he must arrest him since he recognized the pen that belonged to Johnny. However, the criminal recalls that Barney owes him $1,000 that Barney lost in a card game. Barney unsuccessfully tries to raise the money to quit the debt. He decides to tell the chief of police, but out of the blue he sees the spotlight on "The Clarion Call" and runs to the newspaper. What did he see? 3) "The Last Leaf": When 21-year-old Joanna Goodwin's lover breaks up with her, she wanders out into the snow and gets pneumonia. Her older sister Susan discovers that Joanna does not want to live anymore and is following the leaves that keep falling from a tree. Their upstairs neighbor, a painter named Behrman, tries to help the girls. Will he be able to save Joanna? 4) "The Ransom of Red Chief": The con men Sam 'Slick' Brown and Bill Peoria flee to the countryside in their car and plot to kidnap little J.B. Dorset to ask for ransom from his parents, but they soon find that the boy is a little devil. 5) "The Gift of the Magi": On Christmas Eve, Della and her beloved husband Jim are penniless and in love with each other. Jim dreams of giving a tiara to Della since she has wonderful hair, and Della wants to give Jim a chain for his pocket watch. On Christmas night, they each find a way to buy the gifts.

The Donahues - husband and wife Terry and Molly, and their three offspring Steve, Katy and Tim - are a song and dance act. Their survival as a performing act of five and as a family collective is presented. Under their family name, Terry and Molly were a successful vaudeville act in the early 1920s, they who subsequently under the names the Three Donahues, the Four Donahues and the Five Donahues, trotted out Steve, then Steve and Katy, then Steve, Katy and Tim on stage as early as they being toddlers. Molly was able to convince Terry to give the kids a stable education at a boarding school as the two of them continued their on the road career in Molly wanting the kids to have a normal life. They were pleasantly surprised that the kids grew up not only to have musical performing talent, but wanted to perform as a family unit as the Five Donahues. That harmony on and off stage was threatened first by Steve contemplating following another calling - the threat not only in his thought of leaving the act, but in the nature of that calling - and second when Tim becomes infatuated with up and coming singer Vicky Parker, who unlike the naturally talented Donahues, has to tailor her act to showcase her assets. While Vicky is not a bad person per se despite Molly's thoughts to the contrary, Vicky's drive to become a star with Tim's addictive nature proves to be a potentially destructive combination.

During the Korean War, the lieutenant in charge of a Marine rifle platoon is killed in battle. Before he dies, he places the platoon's sergeant, who's black, in charge. The sergeant figures on having trouble with two men in his platoon: a private who has much more combat experience than he does, and a racist Southerner who doesn't like blacks in the first place and has no intention of taking orders from one.

Unfortunately the more naive Mary Lou is impressed by Vanning's oily veneer of materialism and accepts invitations to "entertain" at the gangster's private parties. Mary's naive younger sister Betty arrives from college just when Mary and her roommates are arrested as material witnesses in the murder of one of the casino's non-paying customers. Vanning's corrupt lawyer frees the others but pressures Mary to commit perjury in order to discredit crusading District Attorney David Graham. Disillusioned by her sister's lifestyle. Betty consents to accompany Emmy Lou to one of Vanning's private parties where the gangster ends up killing her for not submitting to the advances of one of the party's guests. Mary decides to tell the truth to the D.A., and is beaten to disfigurement by one of Vanning's hoods. When Steve Emery arrives in Trinidad at the urgent request of his brother, he is stunned to find that his brother has not only been murdered, but that his brother's wife Chris is succumbing to the seduction attempts of the man who quite possibly is the murderer. On board ship, he finds that his direct superior, 'Red' Wildoe, isn't very helpful as far as the galley goes but Al proves to be quite adept at cooking. Al learns a lot about doing the right thing, especially when Stella, now married to Red, starts throwing herself at every man in sight. After the Japanese attack Pearl Harbor, life becomes far more serious.

The respective heads of the Canfield and McKay families try to settle an old score, but by killing each other, they merely perpetuate the old feud. Among the many gags is the train ride that zig-zags around obstacles, a wild chase through the mountains and a hair raising rescue at a waterfall.
