They learn that both suspects had been submitted to a transplant of organs, eye and kidney, and the donor was the same person, the football player Brick Holmes that died in a car crash. They head to Boulder, Colorado, posing as FBI agents and interview Brick's mother Eleanor Holmes that gives vague replies. The Winchester brother seeks out who else received Brick's organs and later they break into his house, where they find a hidden room with a secret.

The Winchester brothers pose as FBI agents to investigate suspicious animal-like killings on a college campus.They question friends MIchael Wheeler, his girlfriend Kate and his friend Brian Wilcox. Wheeler is bitten and turns lycanthropic which seems cool at first and the three spy on Sam and Dean to get more information about what happened to Michael. Jealous of his friends 'superpowers', Brian resolves to be turned too. After Michael refuses to put Brian in danger by biting him, Brian decides to blackmail his professor, Mr. Ludensky--the werewolf that bit Michael. Brian is turned, but the results are horrific and the only thing left behind for Sam and Dean is a video of the devastating events.

The two head back to bayou country to go after Benny's vampire nemesis Quentin, who still holds Benny's true love, Andrea. Benny tries to save Andrea while Dean attempts to eradicate the nest In the meantime. Sam spends time thinking about the life he had while Dean was in Purgatory.

Dean doubts his eyes about seeing Castiel, which re-triggers his purgatory escape trauma, until the archangel simply joins the brothers, claiming amnesia about his escape, but recognizing the abductee names as a list of future prophets, only one of which can be in office at one time. Kevin's mother Linda hires witch Delta Mendota to help make bombs against demons, but her betrayal gets Crowley in, able to kidnap Kevin, whom he tortures to make him read the tablet. The brothers and Castiel find Linda and trail Crowley. Castiel proves himself surprisingly powerful during the fight and reassures Dean, but heaven keeps wiping his memory.

Sam investigates and finds several other victims in earlier suspicious accidents while Dean goes undercover as her bodyguard. Indeed the killer strikes again and proves supernatural.

But it wasn't struck by the in-law they assumed, nor by any of the Cassitys. Dean is tempted by foxy stable hand Ellie. Sam works out why Dean finds it so hard to let him try the triple trial.

Posing as FBI agent, Sam worries, finding James's cop colleague Ed Stoltz seems building up a case against him. Portia's feline colleague Phillippe LeChat is part of a trap, which has the brothers puzzled which side anyone is on, while Sam works on why Dean is so reluctant to let him try the triple challenge from the tablet.

Ajay accepts, but Crowley comes after him once Sam is delivered in purgatory with a portal appointment. Dean gets his demonic mate Benny, who can't get used to honest life anyway, to set up a plan B, which proves desperately needed. Crowley prepares a grim surprise, but gets one from Naomi too.

The trials weakened Sam wearisomely, so after a shooting range test, Dean grounds him to recover. When their hacker acquaintance Charlie Bradbury arrives to report weird killings, Dean grudgingly allows her to come along and clumsily play FBI agent with him, but they can't get passed the stickler coroner and Sam turns up on his own. They figure out it's the work of a blue jinn, whom they find working in the system, and not alone. The only way to counter its feeding on victims' fear is entering dreams, which Sam does with Charlie.

Crowley gets two demons to impersonate the Winchesters to lure paranoid prophet Kevin away, so he would betray the tablet secrets, but is up for a surprise too. Naomi tracks down Castiel despite a clever trick to hide in a fast-food chain, but he hid the tablet gruesomely well. Finding Kevin missing, apparently dead, the brothers start tracking the tablet's origin to an Amerind tribe, only to stumble upon the author, fallen angel 'scribe of God' Metatron.
