While making his daily patrol and thinking about his love life (or the difficulty of getting one), Spider-Man is suddenly surprised by a pair of moments when he seems to lose his Spider-Powers, making him ending inside a trash container full of feathers. In an operation, that involving some kind of "beetle robot" and releasing "The Chameleon" first and a helicopter, Kingpin intends to free from jail a total of six former enemies of Spider-Man: the already mentioned "Chameleon" , "Rhino" , "Doctor Octopus" , " The Scorpion" , "Shocker" and "Mysterio".

Determined to keep his identity safe from the Insidious Six, Spider-Man starts to turn the supervillains against each other, but his powers begin to deteriorate even further. Spider-Man is able to defeat the Insidious Six by telling the villains that he is a fraud. The Insidious Six begins to disband, much to Kingpin's dismay. Spider-Man goes to Dr. Connors so he can do a DNA test on him. Dr. Connors tells Spider-Man that he is going to transform into something that is not human.

After Spider-Man receives some alarming news about his mutation from Dr. Connors, he must contend with the challenge of being a hero until Connors finds a way to cure him. However, Mary Jane receives a call from her former boyfriend, Morris Bench, who has now become Hydro Man. Liz allows Mary Jane to hide at her apartment, but Morris finds Mary Jane and captures her. Morris shows Mary Jane the stolen loot in hopes of winning her over, but she refuses to date him again. Spider-Man is able to defeat Hydro Man, save Mary Jane, and return all of the stolen goods. At an amusement park, Peter and Mary Jane begin to spend some time together, and the couple shares a kiss.

Spider-Man seeks help from Prof. Xavier and his X-Men about the mystery behind his own mutation. Beast advises him to hear Herbert Landon's presentation for a cure on mutation. But unknown to everyone, Landon's new cure is actually meant to kill mutants instead.

Dr. Connors is assigned to study the Tablet of Time, which is said to be able to grant someone their youth back. Dr. Connors shows Spider-Man the tablet and says that he hopes that he can cure Spider-Man of his mutation disease. Before the tablet can be studied properly, both Silvermane and Kingpin want the Tablet of Time for nefarious purposes. Things get more complicated when Spider-Man finds out that Connors has been transforming into the Lizard. At the same time, Peter decides to move on from his relationship with Mary Jane by going out with Alisa Silver, but unbeknownst to him, Alisa is the daughter of the infamous crime lord. Smythe arrives at the Connors' house with his mega-spider slayer, and he captures Margaret. The Lizard escapes, and he starts ambushing Spider-Man in the subway. Spider-Man knocks the Lizard out with an electrical shock from the subway track, and he takes the Lizard back to the lab. Spider-Man is able to change the Lizard back into Dr. Connors, but Tombstone attacks Spider-Man.

Silvermane plans on using the Tablet of Time to make himself young again in hopes of rebuilding his criminal empire. Tombstone has already captured Dr. Connors in order to get him to study the tablet. Spider-Man must rescue Margaret Connors and Alisa Silver from the Kingpin. However, Alisa reveals her true colors to Spider-Man, she knocks out the hero and holds him captive. Spider-Man is shocked that Alisa is the daughter of Silvermane. Dr. Connors tries to plead to Silvermane to not use the tablet but to no avail. Silvermane becomes young again but Connors becomes the Lizard again and he tries to attack Silvermane. However, Silvermane turns into a baby in the process, much to Alisa's fury and dismay. Alisa and Tombstone leave Spider-Man and the Connors for dead, but the hero manages to rescue Curt and Margaret. At the Fisk estate, Vanessa decides to leave her husband after realizing that he is more obsessed with his criminal activities. Kingpin tells Hammerhead to get rid of the tablet. Dr. Connors wakes up at his office and Spider-Man tells him that Silvermane became a baby from the tablet, but they assume the tablet is gone. Unbeknownst to Spider-Man and Dr. Connors, Hammerhead has sold a tablet to an unknown man.

Adrian Toomes provides funding for Dr. Connors and Debra Whitman to work on a upcoming project. Adrian has developed a more sinister agenda when he starts stealing people's youth. Elsewhere, Peter's life hasn't been going great for him, he is still putting up with his mutation disease and he's been receiving a series of treatments from Connors. Peter's relationship with Mary Jane has began to hit an all-time low when she begins to see Harry. Spider-Man must stop Vulture when he begins to set his sights on Norman Osborn, who is planning to take over Adrian's company.

Episode 14

The Vulture has absorbed Spider-Man's youth and powers, but he ends up with Spider-Man's mutated DNA. Spider-Man has turned into an old man and he is unable to perform any heroics. Spider-Man calls Dr. Connors about his predicament and he agrees to help Spider-Man get his youth and powers back from a mutated Vulture. At a mental hospital, Farley Stillwell has been having nightmares of creating the Scorpion and he becomes even more paranoid that the Scorpion will capture him. The Scorpion abducts Stillwell and he takes him to the neogenics lab. Spider-Man arrives at the lab to get Dr. Connors to cure him, but he gets taken down by Scorpion. Dr. Connors reconfigures Vulture's talons, Spider-Man gets his youth and powers back, but Vulture is stuck with the mutated DNA. Farley proceeds to blow up the lab to prevent anymore neogenic experiments. Connors tries to stop Farley, but he becomes the Lizard again. During the ensuing fight, Farley manages to escape the lab and the Lizard soon changes back to his human form. Spider-Man gets Connors to safety, but he is unable to cure himself from his Lizard persona. Farley leaves New York and he goes into hiding. Peter becomes relieved that Connors has freed him of his mutation disease and he goes off to meet Mary Jane, but he discovers that Mary Jane has been captured by an unknown man.
