During his daily patrol, Spider-Man hopes that life in New York City was as peaceful as it is seen from the top. Meanwhile, below the town, two workers face a terrifying creature that manages to capture one of them; the other one was saved by Spider-Man when he put himself and other people in danger while he was driving, still trying to get away from the creature. At the Daily Bugle building, J. Jonah Jameson offers a 1,000 dollars bonus to the reporter who gets a photo of "The lizard". Also, Peter Parker is sent to contact his old teacher and friend, Dr. Curt Connors, to consult him about the subject. Peter (as Spider-Man) manages to take a photo of a giant reptile footprint and a girl Peter knows, named Debra, suggests that the reptile who made the footprint could maybe walk in two legs.

Eddie has become Venom after Spider-Man has successfully discarded the substance at the church bell tower, but Eddie learns that Peter is Spider-Man, he begins to stalk Peter and he threatens to hurt his Aunt May and his girlfriend, Mary Jane. Spider-Man must figure out a way to stop his new adversary before he tries to hurt someone else he cares about.

Norman Osborn and Wilson Fisk (The Kingpin) appear in an event of the Empire State University, Fisk appears as a philanthropist inaugurating what will become the "Wilson Fisk School of Criminology" so that nobody can figure out who he really is. During the ceremony, Peter Parker saves Wilson Fisk of being killed by "The Hobgoblin". After that, Spider-Man and the Hobgoblin have a fight for the first time. Osborn, who had hired the "Hobgoblin" to kill Fisk, recriminates him for not succeeding. The Hobgoblin agrees to go against Kingpin once again after asking double payment. When he is trying to find out more about Kingpin, he is quickly surrounded by his men. When he sees the power of the Kingpin, he is quick to sell his former employer.

While fighting on the streets Hobgoblin thinks he killed Spider-Man but he managed to survive unharmed. Later, the Hobgoblin visits the Kingpin's headquarters. Knowing that Hobgoblin had betrayed him, there is a fight between him and Kingpin's forces and Hobgoblin seems to win, destroying both Kingpin and Smythe. Having survived too, Kingpin and Smythe visit Oscorp and think of an strategy to defeat the Hobgoblin. Osborn tells Spider-Man where he can find the Hobgoblin.

Spider-Man works with a global organization known as S.H.I.E.L.D. which is led by Nick Fury to apprehend an international criminal known as the Chameleon, who is a master of disguise and he plans on ruining a national signing at the U.N. The Chameleon starts to impersonate Fury and Peter in order to throw the web-slinger off his trail. Who can Spidey trust?
